Transcription-mediated amplification (TMA) is an amplification technique carried out by RNA. It uses an isothermal amplification system mediated by RNA transcription and uses two enzymes to drive the reaction: RNA polymerase and reverse transcriptase. Transcription-mediated amplification techniques can be used to target RNA and DNA. TMA has very fast kinetics and can achieve a billion-fold amplification in 15-60 minutes. In addition, it is specific to the target RNA sequence, so it has a wide range of pathogen detection applications in clinical, environmental and food samples.

The best sequence or mutation library selected through high-throughput screening in protein engineering strategies requires a powerful and rapid amplification technology to amplify RNA targets. The transcription-mediated amplification service we provide uses reverse transcriptase to produce DNA from an RNA template, and RNA polymerase to produce RNA from a promoter designed in the primer region. The sensitivity can reach <10 copies of mRNA and <50 copies of rRNA, achieving very rapid production of large amounts of products.

Advantages of Transcription-mediated Amplification

Compared with PCR and LCR amplification systems, TMA-based gene amplification systems has the following other differences:

  • TMA is isothermal. Using a water bath or heating block instead of a thermal cycler.
  • TMA produces RNA amplicons instead of DNA amplicons. Since RNA is less stable than DNA in a laboratory environment, this helps reduce the possibility of residual contamination.
  • Compared to PCR and LCR, which produce only two copies per cycle, TMA produces 100-1000 copies per cycle. This resulted in a 10 billion-fold increase in copies in approximately 15-30 minutes.

Applications of Transcription-mediated Amplification

The polymerase chain reaction gene amplification service is not limited to amplifying cellular mRNA, viral RNA and highly structured ribosomal RNA. The single-stranded RNA amplicons we provide are very suitable for detection probes with oligonucleotides in solution. TMA combined with HPA can specifically perform routine detection and quantification of RNA targets in various samples.
