Usually in living organisms, most protein content is low. To obtain the target protein required in research, it is often necessary to use genetic engineering technology to introduce the nucleic acid sequence encoding the protein into the host cell to express it in large quantities. Prokaryotic expression systems, that is, expressing proteins in bacteria, can usually be divided into E. coli expression systems, Bacillus expression systems, and Streptomyces expression systems. Escherichia coli has the advantages of transparent genetic background, fast cell proliferation, high expression, good stability, and strong anti-pollution ability. Therefore, the E. coli protein expression system is currently the most widely used and most cost-effective bacterial expression system, which is suitable for the expression of various genera of proteins, especially for the production of small molecular proteins. However, there are also problems such as the easy formation of inclusion bodies and endotoxin. Service Content of Protein Expression in Bacterial Cells

  • Gene Synthesis and Codon Optimization. The customer provides the sequence information or plasmid of the target protein. We design the sequence of the appropriate primers according to the customer's experimental requirements, complete the codon optimization and gene synthesis, and finally introduce the target gene into the appropriate vector.
  • Plasmid Transformation and Expression Optimization. The constructed expression plasmid was transferred into host bacteria, the recombinant protein expression was detected, and the optimized expression conditions. We also provide plasmid preparation, plasmid sequencing, and other services.
  • Target Protein Expression and Purification. We can deliver 0.5-1 mg of purified protein samples. If the protein is soluble, we will purify it directly; if it is an inclusion body protein, it will be purified by protein denaturation and renaturation. All purified proteins are verified by rigorous QC (SDS-PAGE, UV, etc.) analysis.
  • Tag-Free Protein Expression and Purification. The amino acid sequence of the tag-free protein is the same as the amino acid sequence of the natural protein, and it is closer to the structure of the natural protein. And it can maintain the natural structure of protein antigens, which is more conducive to protein crystallization experiments and protein function research.
  • Large-Scale Recombinant Protein Production. Our production process is highly optimized for large-scale expression and purification of high-quality recombinant proteins to meet your needs in protein research, protease preparation, drug development, etc.
  • Endotoxin Detection and Removal. Endotoxin can cause fever, microcirculation disturbance, endotoxic shock, and disseminated intravascular coagulation. When it comes to cell and animal experiments, endotoxins need to be tightly controlled. In the prokaryotic protein expression service, bacterial endotoxin removal and endotoxin detection are important parts. We usually use ultrafiltration or affinity chromatography to remove endotoxin and use Limulus reagent to detect endotoxin.
