Introduction of Therapeutic Protein Hormones

Certain diseases characterized by a deficiency or complete loss of endogenous proteins can be treated with alternative therapies that closely mimic the natural pathways of many physiological functions. The most widely recognized protein therapeutics in this field are protein hormones or peptide hormones, including insulin and human growth hormone. More than 60 peptide drugs are currently approved in the US and other major markets. Insulin as a macromolecule has played a prominent role in medical practice since the 1920s. Services

Peptide therapy is limited by the short plasma half-life and low oral bioavailability of native peptides. In addition, the large ligand-binding sites of some peptide GPCRs and the specific conformational changes required for signal transduction pose significant challenges for small-molecule drug discovery.   • Natural peptide improvement: we use modification of natural peptides, analogs and heterologous peptides produced by synthetic or recombinant expression to improve drug properties.
  • Peptide properties: we use conjugation to alter or enhance the properties of peptide and protein drug candidates. Extend the half-life of peptide drugs by conjugation to polyethylene glycol (PEG), lipids and proteins such as Fc fragments.

These strategies we provide allow for modulation of pharmacokinetic properties and target specificity through amino acid or backbone modifications, incorporation of unnatural amino acids, and extension of half-life or improved solubility.
