Cell Surface Displays Service

The cell surface display technology is to introduce the target protein (polypeptide or protein) gene sequence into the cell after fusion with the specific carrier protein gene sequence, so that the target protein is displayed on the surface of the microbial cell. Common carrier proteins include bacterial outer membrane proteins, extracellular accessory structures (such as flagella) proteins, etc. Commonly used host cells include bacteria and yeast cells. Cell surface library sizes of up to 1011 transformants have been reported with each cell displaying thousands of identical copies of a particular mutant protein. High-throughput screening of the library was carried out through a variety of methods, and cells showing proteins with the expected phenotype were Separate for cultivation and reproduction. The characteristics of the carrier protein, target protein and host cell, as well as the fusion method, will affect the efficiency of protein surface display. Advantages of Cell Surface Display System

Although due to the fragility of the cell membrane and other issues, cell display is not as widely used as phage display. But it has the following advantages:

  • The large size of bacteria and yeast cells allows them to be screened using fluorescence microscopy or flow cytometry.
  • The size of the foreign protein to be displayed is quite limited on the surface of the phage. The cell surface display system was developed to solve this problem.
  • Cell surface display allows easy adjustment of library screening conditions, such as buffer composition, pH value, and ionic strength. It usually requires multiple rounds of screening to obtain a rich library of mutants with the desired phenotype.
  • Cell surface display can quickly characterize individual mutants without the need for soluble protein expression and purification.
