The canonical amino acids are limited with respect to their chemical reactivity and the necessity to mutate corresponding residues at other positions to avoid non-specific modification. Non-natural or non-canonical amino acids (NCAAs) acids can now be introduced into recombinant proteins in a residue-specific or site-specific manner, which has major implications for protein engineering. Protein engineering using non-canonical amino acids constitutes a powerful and versatile tool for the redesign of natural proteins and the de novo design of artificial protein-like macromolecules. In recent years, researchers have achieved remarkable results in the application of NCAAs in chemical modification. For instance, NCAAs have been used to form covalent protein–protein and protein–small molecule linkages, creating homogeneous antibody–drug complexes with enhanced effectiveness against target cell types.

Non-canonical amino acids can confer properties not available in the space of classical protein sequences, which can lead to improved or new functions. Creative BioMart has successfully incorporated a variety of non-canonical amino acids into proteins. Our aim is to use non-canonical amino acids to interrogate or modify the reactivity, stability or spectroscopic properties of engineered proteins, designing biomaterials with extraordinary and unique capabilities while increasing protein yields or incorporating multiple/different NCAAs into the same protein.

Strategies of Design Noncanonical Amino Acids Proteins

Complementing the widely used techniques of rational protein design and directed evolution, non-canonical amino acids incorporation has emerged as a new and efficient protein engineering technique. Our scientists have developed a variety of strategies to incorporate NCAAs into proteins for improving the intrinsic function of proteins, creating new possibilities for protein design. For your project, we will choose the right strategy for you.

  • Residue-specific incorporation of noncanonical amino acids: we replace all or part of a specific canonical amino acid with atypical mimetics, typically by using a dystrophic bacterial host to acquire the natural amino acid and supplement the analog.
  • Site-specific incorporation of non-canonical amino acids: we use non-amber nonsense codons to enable site-specific incorporation of unnatural amino acids.
  • New methodology: we develop multiple novel approaches to incorporate non-standard amino acids into proteins, including the design and engineering of new aminoacyl tRNA synthetases and synthetase/tRNA pairs, and the development of ribosomes with altered decoding properties. In addition, we also considered addressing the need for new coding schemes to accommodate standard and non-standard amino acids.
