Protein therapeutics according to its molecular type, including antibody-based drugs, anticoagulants, blood factors, bone morphogenetic proteins, engineered protein scaffolds, enzymes, Fc fusion proteins, growth factors, hormones, interferons, interleukins and thrombolytic drugs, etc. Protein drugs are usually large in size and have a larger surface area to contact the target protein, making them an indispensable therapeutic tool for the treatment of human diseases.

These proteins can achieve desired properties through specific mutations, fusion partners, or chemical modifications. Strategies of Engineering of Therapeutic Proteins

Protein therapeutics needs a lot of improvement in drug target identification, protein engineering and design, protein expression and purification, and drug delivery. In order to solve these problems, our scientists are committed to developing various strategies for designing effective protein therapeutics.   • Strategies for improving pharmacokinetics: we use a variety of strategies including fusion, glycosylation, and chemical modification to improve the efficacy of therapeutic proteins in the human body. Recombinant DNA technology can fuse the protein of interest with endogenous human proteins, thereby increasing the effective size of the protein and reducing the clearance rate in the kidney. Glycosylation helps enhance the solubility of proteins. Polyethylene glycol (PEG) modification increases the half-life of serum proteins.
  • Strategies for reducing immunogenicity: we have developed various strategies to reduce immunogenicity. Polyethylene glycol (PEG) protects the immune response sites on the recombinant protein from being recognized by antibodies or surface receptors. Changing the main sequence of the therapeutic protein can also help reduce immunogenicity. In addition, recognition and elimination of antibodies and T-cell epitopes and class II MHC agretopes are used to reduce the immunogenicity of next-generation protein therapies.
  • Genetic Engineering Strategies: we use the three strategies of rational design, directed evolution and semi-rational design to change the physical and chemical properties of proteins and/or create new functions, and apply them to protein therapeutics.
