Ribosome Displays Service

The ribosome display system is an effective in vitro cell-free system. It uses functional protein interactions for screening and binds the correctly folded protein and its mRNA to the ribosome at the same time to form an mRNA-ribosome-protein trimer. Ribosome display technology links the genotype and phenotype of the target protein, and can be used for antibody and protein library selection, protein in vitro modification, etc.

Creative BioMart provides a unique ribosome display platform based on our proprietary prokaryotic, eukaryotic ribosome display and ribosome inactivation display technology. Our ribosome display technology is carried out completely in vitro, which makes up for the shortcomings of phage display and cell surface display. Therefore, it can significantly increase the storage capacity and molecular diversity, among which the storage capacity can reach 1013 to 1014, which can be used in the screening and research of any cytotoxic molecules. Prokaryotic ribosome display system service

Prokaryotic ribosomes are shown to use the coupled E. coli S30 system for peptide selection. Creative BioMart provides E. coli S30 extract display system. This system demonstrates the selection of peptide ligands similar to the known peptide epitopes of a given antibody, using that antibody as a selection substrate.

Eukaryotic ribosome display system service

The eukaryotic ribosome display system is used to select functional single-chain antibody fragments in the coupled rabbit reticulocyte lysis system. The rabbit reticulocyte lysate display system provided by Creative BioMart adopts an in-situ recovery program, in which a new in-situ RT-PCR program is used for amplification, and DNA is directly recovered on the ribosome complex without dissociating the ribosome complex.

Ribosome inactivation display system (RIDS) service

RIDS is a powerful peptide and protein selection and evolution technology. The RIDS provided by Creative BioMart shows that the ricin A chain is used in the system. Once the ricin A chain completely emerges from the ribosome, it will catalyze a single hydrolysis event in the large ribosome subunit, thereby changing changes the binding site of the elongation factor and preventing the complex.

The ribosome display service we provide is widely used to select proteins and peptides in vitro from large libraries. It can display very large libraries, is suitable for the production of toxic, proteolytically sensitive and unstable proteins, and also allows amino acid modifications at specific positions.

Features of Ribosome Display System

  • The process of ribosome display does not require a complicated separation process, and provides a redy-made method to amplify indeed bound proteins without the need to decrypt the sequence until necessary.
  • Ribosome display technology relies on generating a large and concentrated sequence diversity library without gaps, and preventing these sequences from degrading, hybridizing, and interacting in a way that would create gaps in sequence space.
  • Compared with other display technologies, the diversity of ribosome display libraries is not limited by the transformation efficiency of bacterial cells, but only by the number of ribosomes present in the test tube and different mRNA molecules. Second, random mutations can be easily introduced into the selected DNA library in subsequent cycles.
  • In ribosome display, the ribosome complex is allowed to bind to a target immobilized on the surface. The unbound complex is washed away, and the mRNA displaying the complex bound to the polypeptide can be recovered. Therefore, the genetic information of the bound polypeptide can be used for analysis.
